Tyre size and pressure

Maybe I'm wording this wrong but google has not provided me with an answer.

I want to know if changing to larger wheels with wider lower profile tyres will affect the tyre pressure? Or does it stay the same?

Also a slightly off topic question - lets say there are no people in a car and all four tyres are at 36psi. Will the tyre pressure still measure 36psi with four blokes in it or is it going to change?
